Chromosomal origins of DNA replication in higher eukaryotes differ significantly from those of E. coli (oriC) and the tumor virus, SV40 (ori sequence). Initiation events appear to occur throughout broad zones rather than at specific origin sequences. Analysis of four chromosomal origin regions reveals that they share common modular sequence elements. These include DNA unwinding elements, pyrimidine tracts that may serve as strong DNA polymerase‐primase start sites, scaffold associated regions, transcriptional regulatory sequences, and, possibly, initiator protein binding sites and inherently (...) destabilized regions. Based on the novel organization of chromosomal origin regions, we propose a model for initiation of DNA replication in higher eukaryotes. Unwinding of duplex DNA during initiation may be uncoupled, both temporally and spatially, from DNA synthesis, resulting in transient single‐stranded intermediates that function in lieu of conventional replication forks during chromosomal DNA replication. Argumentation as the public exchange of reasons is widely thought to enhance deliberative interactions that generate and justify reasonable public policies. Adopting an argumentation-theoretic perspective, we survey the norms that should govern public argumentation and address some of the complexities that scholarly treatments have identified. Our focus is on norms associated with the ideals of correctness and participation as sources of a politically legitimate deliberative outcome. In principle, both ideals are mutually coherent. If the information needed for a correct deliberative (...) outcome is distributed among agents, then maximising participation increases information diversity. But both ideals can also be in tension. If participants lack competence or are prone to biases, a correct deliberative outcome requires limiting participation. The central question for public argumentation, therefore, is how to strike a balance between both ideals. Rather than advocating a preferred normative framework, our main purpose is to illustrate the complexity of this theme. (shrink)

Due to the COVID-19 pandemic, cities and states adopted social distancing, social isolation, or quarantine measurements to slow the transmission of the disease. Negative mental health outcomes including depression and anxiety have been associated with social distancing or social isolation. The purpose of the present study was to examine changes in psychological health and physical activity over an 8 week period under social distancing policies during the COVID-19 pandemic.Methods: Ninety individuals participated in this study. Qualifying participants answered questions using an (...) online survey regarding their loneliness, depressive symptoms, anxiety symptoms, mood state, and physical activity over four time points each lasting two weeks.Results: Symptoms of depression and state anxiety were increased in the population when compared to nationwide statistics from before the COVID-19 pandemic. Time point 2, ~1 month into social isolation, showed the most significant effects on mental health. During this time point, 100% of the participants showed symptoms of depression. There were no significant changes in physical activity over the 8 weeks. Loneliness, depressive symptoms, fatigue, and mood state were negatively associated with participation in physical activity. Vigor and state anxiety were associated with participation in physical activity.Conclusion: Social isolation and social distancing practices have had a negative effect on depression, anxiety, and mood over time. It appeared that depressive symptoms and total mood disturbance was elevated during time point two. Depressive symptoms were much higher than average compared to previous epidemiological data. Physical activity amount did not change over time but was associated with poor mental health. (shrink)

Preadolescence is an important period for the consolidation of certain arithmetic facts, and the development of problem-solving strategies. Obese subjects seem to have poorer academic performance in math than their normal-weight peers, suggesting a negative effect of obesity on math skills in critical developmental periods. To test this hypothesis, event-related potentials were collected during a delayed-verification math task using simple addition and subtraction problems in obese [above 95th body mass index percentile] and non-obese preteens with different levels of math skill; (...) thirty-one with low math skills and thirty-one with high math skills. No significant differences between weight groups were observed in task accuracy regardless of their mathematical skill level. For ERPs, electrophysiological differences were found only in the subtraction condition; participants with obesity showed an electrophysiologic pattern associated with a reduced ability to allocate attention resources regardless of their math skill level, these differences were characterized by longer P300 latency than their normal-weight peers. Moreover, the participants with obesity with high math skills displayed hypoactivity in left superior parietal lobule compared with their normal-weight peers. Additionally, obese preteens with low math skills displayed smaller arithmetic N400 amplitude than non-obese participants, reflecting difficulties in retrieving visual, semantic, and lexical information about numbers. We conclude that participants with obesity are less able than their normal-weight peers to deploy their attention regardless of their behavioral performance, which seems to have a greater effect on obese participants with low math skills because they also show problems in the retrieval of solutions from working memory, resulting in a delay in the development of mathematical skills. In any attempt to present an overview of the conceptions of reality and divinity in classical Indian (Hindu) civilization, it is helpful, first of all, to highlight some of the basic cultural and intellectual presuppositions that appear to be operative in classical Indian thought (which, for the purposes of this article, will be taken as consisting of the so‐called six classical schools of Sāṃkhya, Yoga, Nyāya, Vaiśeṣika, Mīmāṃsā, and Advaita Vedānta during the classical period, from the first centuries of the (...) Common Era up to the coming of Islamic traditions in the eleventh century). This can be accomplished in various ways, but perhaps the simplest way is to articulate some basic questions to which classical Indian thought provides some interesting, albeit unusual, answers. The questions can be framed as follows: (1) What truly is? (2) How does one know what truly is? (3) Who knows what truly is? (4) With whom does one know what truly is? And, finally: (5) What does God have to do with what truly is? The questions have to do, respectively, with (1) ontology, (2) epistemology, (3) psychology, (4) social anthropology, and (5) theology. Classical Indian thought, of course, provides a variety of answers to all of these questions, and there was much vigorous debate and polemic through the centuries about precisely how to form satisfactory answers. Generally speaking, however, it appears to be the case that the classical Indian philosophical schools tended to deal with these basic questions in a manner quite different from the way these questions have been dealt with in traditional or modern Western thought, and it is important to understand some of these differences if one wishes to appreciate the unique conceptual frameworks in classical Indian thought. The Filter Dichotomy says that every uniform nonmeager filter on the integers is mapped by a finite-to-one function to an ultrafilter. The consistency of this principle was proved by Blass and Laflamme. A medial limit is a universally measurable function from [Formula: see text] to the unit interval [0, 1] which is finitely additive for disjoint sets, and maps singletons to 0 and ω to 1. The forcing construction Pmax, invented by W. Hugh Woodin, produces a model whose collection of subsets of ω₁ is in some sense maximal. In this paper we study the Boolean algebra induced by the nonstationary ideal on ω₁ in this model. Among other things we show that the induced quotient does not have a simply definable form. We also prove several results about saturation properties of the ideal in this extension.

This chapter explores the debate between contemporary projectivists or expressivists, and the advocates of sensibility theory. Both positions are best viewed as forms of sentimentalism — the theory that evaluative concepts must be explicated by appeal to the sentiments. It argues that the sophisticated interpretation of such notions as “true” and “objective” that are offered by defenders of these competing views ultimately undermines the significance of their meta-ethical disputes over “cognitivism” and “realism” about value. The authors present classical and recent results of linguistic semantics within the framework of interpretative T-theories and defend the philosophical foundations of their approach by showing how it fits into the larger enterprise of cognitive linguistics. Traditionally, philosophers have ascribed moral agency almost exclusively to humans. Early writing about moral agency can be traced to Aristotle and Aquinas. In addition to human moral agents, Aristotle discussed the possibility of moral agency of the Greek gods and Aquinas discussed the possibility of moral agency of angels. In the case of angels, a difficulty in ascribing moral agency was that it was suspected that angels did not have enough independence from God to ascribe to the angels genuine moral (...) choices. Recently, new candidates have been suggested for non‐human moral agency. Floridi and Sanders suggest that artificially intelligence programs that meet certain criteria may attain the status of moral agents; they suggest a redefinition of moral agency to clarify the relationship between artificial and human agents. Other philosophers, as well as scholars in Science and Technology Studies, are studying the possibility that artifacts that are not designed to mimic human intelligence still embody a kind of moral agency. For example, there has been a lively discussion about the moral intent and the consequential effects of speed bumps. The connections and distributed intelligence of a network is another candidate being considered for moral agency. These philosophical arguments may have practical consequences for software developers, and for the people affected by computing.
